Two people travelling in the Ford Focus have been taken to hospital with minor injuries.
Sergeant Meurig Jones of the Roads Policing Unit said: “Sadly two people have died and our thoughts very much remain with their family and friends.
“The motorcycle – which is a foreign-registered vehicle, was travelling in a group of four motorcycles from the direction of Denbigh towards Groes. The Ford Focus was travelling in the opposite direction.
“We are appealing for any witnesses who may have seen the bikes prior to the collision to come forward.
"We are also appealing to anybody who may have been travelling in the area at the time and who may have dash cam footage to contact us.”
The A543 remains closed in that area and officers are asking people to avoid the area and seek alternative routes.
Anybody with information is asked to contact officers at the Roads Policing Unit on 101 or via the live web chat quoting reference number W050328.
